0 hours: RNA-seq was performed under sporulation (meiotic induction) conditions. Overnight YPD cultures were diluted to OD600 of 0.1 in YPA (1% yeast extract, 2% peptone, 1% potassium acetate) and grown 16-18 h at 30°C. Cells were then resuspended at OD600 of 0.5 in 1% potassium acetate (Spo medium) supplemented with uracil, histidine, and leucine, and incubated at 26°C. RNA-seq samples were collected at 0 hours post-transfer to Spo medium.
